Using the Pomeroy, Salisbury Twp, Meigs Co., OH, 1850 census as a baseline, Joseph Isenhour's family, #242-250, consists of wife, Lydia, daughters M.S. (Mary) b. VA abt 1843, Emeline b. MI abt 1845, and Caroline b. OH abt 1849. The birth information for the children is relatively constant through the 1870 census. In the 1860 and 1870 censuses Sarah Hart b. IA abt 1857 is included in the family. Lydia disppears after the 1870 census, as does Sarah, and Joseph is living with his daughter Caroline and her family in the 1880 census. Mary and Emma married before 1880, raised children, and remained in Meigs Co., Ohio.
In May 1842, Joseph married Lydia Caylor in Mason, VA, later West Virginia. In the four censuses in which Lydia appears, twice her birth state is given as Pennsylvania and twice as Ohio. It's likely that their first daughter, Mary, was born in Mason County, and the Virginia appearing on census records as her birth state is pre Civil War Virginia.
